How does Survodutide work?

How is Survodutide administered?

What does Survodutide stack well with?

Survodutide's dual-agonist mechanism is potent on its own, and stacking it with other GLP-1-class compounds tends to amplify gastrointestinal side effects without proportional benefit. The cleanest additions are non-peptide: resistance training, adequate protein, and — given the liver-directed mechanism — attention to alcohol and hepatotoxic exposures. The items to avoid overlap the same incretin pathways.

  1. Standalone use. The most common research protocol. The dual-agonist effect is strong enough that adding other GLP-1-class peptides is generally unnecessary and compounds side effects.
  2. Resistance training + 1.2–1.6 g/kg protein. Used to preserve lean mass during rapid weight loss, as with all incretin-class agents.
  3. Liver-supportive habits. Because the glucagon arm acts on the liver, minimizing alcohol and other hepatic stressors is a logical accompaniment in research contexts.
  4. Avoid: other GLP-1-class agonists. Combining survodutide with semaglutide, tirzepatide, retatrutide, or mazdutide stacks the same receptor pathways and sharply increases GI side effects without added benefit.

What are the side effects of Survodutide?

Survodutide's side-effect profile is dominated by gastrointestinal effects, consistent with the GLP-1 class, and these are most pronounced during dose escalation. Glucagon and GLP-1 receptor dual agonist survodutide for obesity: a randomised, double-blind, placebo-controlled, dose-finding phase 2 trial

The Phase 2 dose-finding obesity trial of survodutide in 386 adults (survodutide n=309; placebo n=77) with overweight or obesity. After a 20-week dose-escalation and 26-week maintenance period, mean body weight change to week 46 under the primary treatment-policy analysis was −6.2% (0.6 mg), −12.5% (2.4 mg), −13.2% (3.6 mg), and −14.9% (4.8 mg), versus −2.8% on placebo. Up to 40% of participants reaching the two highest doses lost at least 20% of body weight. Gastrointestinal effects were the most common adverse events and were mitigated by slow titration. This established survodutide's dose-response and set the doses carried into the Phase 3 SYNCHRONIZE obesity program.

A Phase 2 Randomized Trial of Survodutide in MASH and Fibrosis
